- Balloon Bash mode: Reach Level 3.
- Boo’s Block Party mode: Reach Level 9.
- Challenge Tower mode: Reach Level 10.
- Coinathlon mode: Reach Level 2.
- Dr. Mario Music Rhythm Recital level: Reach Level 11.
- Mario Shuffle mode: Reach Level 7.
- Master difficulty: Reach Level 12.
- Master Tower (Challenge Tower): Successfully complete the Expert Tower.
- Percussion Set (Rhythm Recital): Get an “A” rank or higher on at least five different songs.
- Rhythm Recital mode: Reach Level 5.
- Staff Credits: Reach Level Star.
- Star Rush Rank (Toad Scramble/Balloon Bash): Fill up all the star slots in the result screen of a board.
- Super Star Rush Rank (Toad Scramble/Balloon Bash): Fill up all the star slots and more in the result screen of a board.
- Tower Cup (Challenge Tower): Successfully complete the Master Tower.
- Unlock Diddy Kong: Reach Level 11 or scan a Diddy Kong Amiibo.
- Unlock Donkey Kong: Reach Level 8 or scan a Donkey Kong Amiibo.
- Unlock Rosalina: Reach Level 6 or scan a Rosalina Amiibo.
- Unlock Toadette: Reach Level 4.
